I suggest the coastal route going North, visiting Padang Bai.
At Amlapura, go past the water palace at Ujung, and onto the spectacular hilly and windy coastal road.
This gives access to the Amed strip of villages, entering from the south.
When leaving Amed, continue north and work your way towards the west and Tirtaganga water palace.
You can then return south having made a "circle" around the eastern part of the Island and not backtracking.
Personally, I think it deserves at least a day, with a half way stop somewhere like The Puri Raja.....but that's just my view.
