there are bemos going to the places you mention from Padang Bai. They leave from different spots, ask as your accommodation. There will also be plenty of drivers available so it will be a personal decison.
tenganan village is not on the bemo route, you would be dropped off on the main road- its about a 3km walk along the road into tenganan -there will also be ojeks (motorbikes) who will take you up.
I would actually do a day trip to tenganan and klungkung with a driver (maybe ask around fellow travellers to share with), can easily cover both in a day, then if you like culture and scenery stay for a night or two in Tirtagganga. There are beautiful walks or bike rides through the terraces and villages and you can spend as much tiem as you like at the waterpalace.
