There are actually a lot of buses doing this trip on a daily basis. Once you are in Sanur there are many small places along Jalan Tamblingan that sell tickets for various prices. The buses range from local "pack em in" type up (or down) to small 20 seater airconditioned buses.
You will have very little trouble getting a ticket, except around the Ramadan period when many of the Javanese Muslims head home.
